What is confluent growth on a bacterial plate?

50% confluence means half of the area is covered by bacterial cells. If the growth of the bacteria covers almost the entire area of the membrane filter or culture media like agar then it is known as confluent growth. Say in the streak plate method, the initial region of inoculation shows confluent growth.

How do you create a bacterial culture?

Before you can grow bacteria, you'll need to prepare sterile culture dishes. A 125ml bottle of nutrient agar contains enough to fill about 10 petri dishes. Water Bath Method – Loosen the agar bottle cap, but do not remove it completely. Place the bottle in hot water at 170-190 °F until all of the agar is liquid.

How do bacteria spread on agar plates?

Procedure of Spread Plate Technique

Make a dilution series from a sample.
Pipette out 0.1 ml from the appropriate desired dilution series onto the center of the surface of an agar plate.
Dip the L-shaped glass spreader into alcohol.
Flame the glass spreader (hockey stick) over a Bunsen burner.
Spread the sample evenly over the surface of agar using the sterile glass spreader, carefully rotating the Petridish underneath at the same time.
Incubate the plate at 37°C for 24 hours.
Calculate the CFU value of the sample. Once you count the colonies, multiply by the appropriate dilution factor to determine the number of CFU/mL in the original sample.
